What Windows version are you using? You should be able to open the .iso by right clicking and choosing 'mount' - in the list of attached devices it looks as though there's a DVD drive with the disc in it, even though there isn't a physical drive on your computer (this is called a virtual drive).
I often use an ancient piece of software called DVD Decrypter to extract the movie's .vob as a single file rather than making an .mkv, but whatever works for you. Once the .iso is mounted you can do anything you'd do with a physical DVD inserted into a real drive. To un-mount the disc image when you're done you click on the virtual drive and choose Eject. It then disappears from the list. Unless you eject it, you won't be able to move or delete the .iso because the file is in use, like an open document. Gotta close the file first!
Another way is to simply open the .iso using WinRAR or 7Zip, as Ghost suggests. Then extract the contents to a folder on your hard drive and go from there.

I've been using Makemkv for a long time, I already extracted the film yesterday and I'm preparing the French subtitles...

The subject may have been broached, but does anyone know why Fabrizio's final confession in the subtitle is displayed in silence when there is no voice-over ? Why this important information (otherwise we don't know that Silvia survives) is not given in text embedded on the screen like the final poem ?

I think the final confession is only in one of the dubbed languages but not the other(s), and the subtitle matches that one.
